Clerodendrum wallichii Merr is a tall ornamental bushy shrub native to southern Asia and Southern China. Traditionally, the leaves of the plant are used for the treatment of fever, skin infections, inflammation, diarrhea and dysentery. Pharmacological and biological investigations on Clerodendrum wallichii is scarce and no scientific investigation has been conducted to evaluate its effect on liver. We, therefore, aimed to evaluate the protective effect of Clerodendrum wallichii methanol extract (MECW) against alcohol-induced liver injury in animal model. Oral acute toxicity study was carried out according to OECD guideline. The hepatotoxicity was induced by continuous administration of ethanol for 4 weeks. The hepatic enzymes (SGPT, SGOT, and ALP) and oxidative stress biomarkers (CAT, GST, SOD, MDA and H2O2) were measured using commercial diagnostic kits. Treatment of rats with MECW (800 mg/kg) showed the highest reduction of body weight (4.76±0.372 vs. 16.92±0.846) and liver weight (3.06±0.128 vs 5.55±0.311). Treatment of alcohol-induced rats with MECW at 200, 400, 600, 800 & 1000 mg/kg significantly (**p<0.01) reduced the level of SGPT. Similarly, serum SGOT and ALP were significantly decreased by MECW doses of 200, 400, 600, 800 and 1000 mg/kg. Treatment of rats with all the doses of MECW significantly increased anti-oxidant enzymes GSH and SOD. Similarly, MECW at doses of 600 and 800 mg/kg significantly promoted CAT levels in liver tissues. On the contrary, MECW treatment significantly diminished oxidative biomarker, MDA. Histopathological observations of liver showed improvement of architecture of hepatic cells having the signs of protection with reduced number of inflammatory cells, vascular degeneration and congestion, cellular degeneration, necrosis and significance reduction of fatty cells accumulation. Acute toxicity study resulted with well-tolerability and safety of MECW doses (200-1000 mg/kg) in laboratory rats. Our study, thus, clearly demonstrated the hepatoprotective effect of Clerodendrum wallichii extract against alcohol-induced liver injury in animal model.
